To the stirred solution of tetrahydro-4H-pyran-4-one (2 g, 9.33 mmol), tert-butoxycarbonyl hydrazide (1.8 g, 13.99 mmol) in AcOH: MeOH (1 :1 , 10 mL), NaCNBH3 (0.52 g, 8.4 mmol) was added portion wise at 0 C and the reaction mixture was stirred at RT for 30 min. Completion of the reaction was monitored by TLC; the reaction mixture was then concentrated under vacuum. The resulting mixture was suspended with 10% NaHC03 (20 mL) and was extracted with DCM (2 x 15 ml_). The combined organic layer was washed with brine (10 mL), dried over anhydrous Na2S04 and concentrated under vacuum to afford the title compound. Yield: 99% (crude) (2 g, off white solid). 1H NMR (400 MHz, DMSO-d6): delta 8.20 (s, 1 H), 4.34 (s, 1 H), 3.82-3.81 (m, 2H), 3.29-3.26 (m, 2H), 2.89-2.88 (m, 1 H), 1.63 (t, J = 2.0 Hz, 2H), 1.38 (s, 9H), 1.24 (t, J = 4.0 Hz, 2H).
