Example 96A 6,7-dihydro-4H-pyrano[3,4-d]thiazol-2-amine To a solution of lithium diisopropylamide (2.5 mL, 5.0 mmol, 2M in THF, Aldrich) in THF (20 mL) was added drop wise dihydro-2H-pyran-3(4H)-one (0.5 g, 5 mmol, Small Molecules Inc) in THF (2 mL) at -78 C. The reaction mixture was stirred at -45 C. for 2 hr and then cannulated to a solution of sulfur (0.16 g, 5.0 mmol) in THF (20 mL) at -45 C. The reaction mixture was warmed to 0 C. and a solution of cyanamide (0.42 g, 10.0 mmol) in THF (2 mL) was added. After stirring for overnight, the reaction mixture was quenched with saturated NaHCO3 (10 mL). The aqueous layer was extracted with ethyl acetate (3*20 mL). The combined organic extracts were dried (Na2SO4), filtered and concentrated. The residue was purified by column chromatography using an Analogix Intelliflash280 (SiO2, 0-5% methanol in dichloromethane) to afford 0.1 g (10%) of the title compound.
