125552-89-8, 4-(Bromomethyl)tetrahydropyran (797 mg; 4.45 mmol) is added to a mixture of 3-oxo-3,4-dihydro-2H-benzo[1,4]thiazine-7-sulfonic acid (4-ethylphenyl)isobutylamide (900 mg; 2.22 mmol) and cesium carbonate (1.09 g; 3.34 mmol) in 1-methyl-2-pyrrolidone (20 ml). (0657) The reaction medium is stirred for 4 hours at a temperature of 110 C., hydrolyzed and extracted with ethyl acetate. The organic phases are combined, washed with brine, dried (Na2SO4) and concentrated. (0658) The crude product is chromatographed on silica gel (eluent: heptane/ethyl acetate, from 0 to 60% of ethyl acetate). The 3-oxo-4-(tetrahydropyran-4-ylmethyl)-3,4-dihydro-2H-benzo[1,4]thiazine-7-sulfonic acid (4-ethylphenyl)isobutylamide (1.11 g; 99%) is obtained in the form of a white solid with a compliant 1H NMR.
