Purification and Characterization of N-Acetyl-D-hexosamine Dehydrogenase from Pseudomonas sp. No. 53

A new enzyme, N-acetyl-D-hexosamine dehydrogenase (N-acetyl-alpha-D-hexosamine: NAD+ 1-oxidoreductase), was purified to homogeneity on polyacrylamide gel electrophoresis from a strain of Pseudomonas sp. about 900-fold with a yield of 12percent.The molecular weight of the enzyme was about 124,000 on gel filtration and 30,000 on SDS-polyacrylamide gel electrophoresis, respectively.Its isoelectric point was 4.7.The optimum pH was about 10.0.The enzyme was most stable between pH 8.0 and pH 10.5.The highest enzyme activity was observed with N-acetyl-D-glucosamine (Km=5.3 mM) and N-acetyl-D-galactosamine (Km=0.8 mM) as the sugar substrate.But it was not so active on N-acetyl-D-mannosamine.NAD+ was used specifically as the hydrogen acceptor.The anomeric requirement of the enzyme for N-acetyl-D-glucosamine was the alpha-pyranose form, and the reaction product was N-acetyl-D-glucosaminic acid.The enzyme activity was inhibited by Hg2+ and SDS, but many divalent cations, metal-chelating reagents, and sulfhydryl reagents had no effect.

The synthesis and spectroscopic investigation of the new noncyclic neutral ligands 12-20 of the glycol ether type (podands) carrying terminal electron-donor- and -acceptor groups are described.The podands show intramolecular charge transfer interactions which can be influenced selectively by complexed cations.The ligands 13 and 20 cause significant increases of extinction and bathochromic shifts of the charge transfer bands in the UV/Vis spectra on addition of alkali perchlorates, whereas the shorter ligands 14 and 18 show a decrease of extinction and hypsochromic shifts under similar conditions.Some of the noncyclic chromoionophores discern considerably and visually perceivably between Na+ and K+ or Li+ and Na+, respectively, in acetonitrile.The cation selective alterations in the electron spectra are discussed in view of steric effects considering some complex stability constants determined by photometric titrations.

Design and Synthesis of Natural Product Inspired Libraries Based on the Three-Dimensional (3D) Cedrane Scaffold: Toward the Exploration of 3D Biological Space

A chemoinformatic method was developed to extract nonflat scaffolds embedded in natural products within the Dictionary of Natural Products (DNP). The cedrane scaffold was then chosen as an example of a nonflat scaffold that directs substituents in three-dimensional (3D) space. A cedrane scaffold that has three orthogonal handles to allow generation of 1D, 2D, and 3D libraries was synthesized on a large scale. These libraries would cover more than 50% of the natural diversity of natural products with an embedded cedrane scaffold. Synthesis of three focused natural product-like libraries based on the 3D cedrane scaffold was achieved. A phenotypic assay was used to test the biological profile of synthesized compounds against normal and Parkinson’s patient-derived cells. The cytological profiles of the synthesized analogues based on the cedrane scaffold revealed that this 3D scaffold, prevalidated by nature, can interact with biological systems as it displayed various effects against normal and Parkinson’s patient-derived cell lines.
