A clinician’s guide to statin drug-drug interactions

The statins are widely used worldwide to reduce risk for cardiovascular events in both the primary and secondary prevention settings. Although generally quite safe, the statins can be associated with a variety of serious side adverse effects, including myalgia, myopathy, and changes in plasma enzymes of hepatic origin. Although rare, the most serious of these is rhabdomyolysis. Several drugs can interfere with the metabolism and disposal of the statins, thereby increasing risk for adverse events. It is important that clinicians treating patients with statins be aware of the potential for drug-drug interactions between each statin and specific other drugs and take measures to prevent them. The prediction of potential drug-drug interactions derives from basic pharmacokinetic principles. Certain drug interactions are predicted by measuring the effect of interacting drugs on blood plasma concentrations of the statin. Individual patient variations resulting in part from polymorphisms in the metabolizing enzymes confound some of these predictions. Based on these known effects, a new classification for predicting statin drug interactions is proposed. This report discusses likely prescription and nonprescription interactions as well as potential alternatives for special populations. Regioselective 4,6-O-arylidenation of glycosides and thioglycosides with benzaldehyde or p-methoxybenzaldehyde dimethyl acetal is catalyzed by 10 mole% of Mg(OTf)2 to produce the corresponding 4,6-O-arylidenated product in high yields. Mg(OTf)2 can also mediate sequential one-pot benzylidenation-acetylation of mono and disaccharide based glycosides and thioglycosides in high yield.

Two new ortho-metalated rhodium(iii) complexes of the formula [Rh(ppy) 2(L)]+, ppy = 2-phenylpyridine and L = 2,2?-dipyridylketone (dpk) (1), 2,2?-dipyridylamine (HDPA) (2) have been synthesized and subjected to X-ray diffraction crystal structural, photophysical and electrochemical studies. Density functional theory calculations have also been performed to get rationalizations of the optical orbitals and redox orbitals concerning photophysical and electrochemical data. Complex 1 exhibits the triplet ligand-to-ligand charge transfer ( 3LLCT) [pi(ppy)-pi*(dpk)] phosphorescence at 77K (520 nm) and at room temperature (555 nm), while complex 2 shows triplet ligand centred (3LC) [pi-pi*(ppy)] phosphorescence only at 77K (460 nm). Both complexes 1 and 2 have similar irreversible oxidation potentials (+1.19 V for 1 and +1.15 V for 2 vs. Fc/Fc+). These two complexes show different characteristics in the reduction process: a reversible process occurs for 1 at -1.31 V, while an irreversible process is observed for 2 at -1.85 V. A robust synthesis of N-glycolyl muramyl dipeptide via azidonitration/reduction

A novel synthetic route leading to N-glycolyl muramyl dipeptide (MDP), a bacterial glycopeptide of particular interest in studies of nucleotide-binding oligomerization domain-containing protein 2 (NOD2), is described. The synthetic strategy hinges on the alkylation of benzylidene-protected glucal with 2-bromopropionic acid and thus circumvents a challenging and non-reproducible SN2 step at the C-3 position of glucosamine derivatives. The subsequent sequence includes an azidonitration and an unusual azide reduction/acylation step via an aza ylide/oxaphospholidine intermediate. This approach generates a protected N-glycolyl MDP that can be either subjected to a one-step global deprotection or differentially deprotected to obtain further derivatives.

Structural analysis and anthelmintic activity of Canavalia brasiliensis lectin reveal molecular correlation between the carbohydrate recognition domain and glycans of Haemonchus contortus

Haemonchus contortus is one of the most economically important parasites infecting small ruminants worldwide. This nematode has shown a great ability to develop resistance to anthelmintic drugs, calling for the development of alternative control approaches. Because lectins recognize and bind to specific carbohydrates and glycan structures present in parasites, they can be considered as an alternative to develop new antiparasitic drugs. Accordingly, this work aimed to investigate the anthelmintic effect of Canavalia brasiliensis (ConBr) lectin against H. contortus and to evaluate a possible interaction of ConBr with glycans of this parasite by molecular docking. ConBr showed significant inhibition of H. contortus larval development with an IC50 of 0.26 mg mL?1. Molecular docking assays revealed that glycans containing the core trimannoside [Man(alpha1-3)Man(alpha1-6)Man] of H. contortus interact in the carbohydrate recognition domain of ConBr with an interaction value of MDS = ?248.77. Our findings suggest that the inhibition of H. contortus larval development is directly related to the recognition of the core trimannoside present in the glycans of these parasites. This work is the first to report on the structure-function relationships of the anthelmintic activity of plant lectins.
