A dendronized carbohydrate thermotropic liquid crystal was synthesized by attaching wedge-shaped mesogens onto a carbohydrate core. These molecules self-organize into chiral columnar hexagonal mesophase with each column slice (4.5 thicknesses) filled with average of two molecules. The supramolecular model was further optimized by molecular dynamics simulation. Moreover, chirality successfully expressed in columnar hexagonal mesophase by dendronized carbohydrate molecules may provide inspiration in searching for chiral mesophase of carbohydrate liquid crystals.

Gram-negative bacteria have evolved an elaborate process for the recycling of their cell wall, which is initiated in the periplasmic space by the action of lytic transglycosylases. The product of this reaction, beta-d-N- acetylglucosamine-(1a?’4)-1,6-anhydro-beta-d-N-acetylmuramyl-l- Ala-I-d-Glu-meso-DAP-d-Ala-d-Ala (compound 1), is internalized to begin the recycling events within the cytoplasm. The first step in the cytoplasmic recycling is catalyzed by the NagZ glycosylase, which cleaves in a hydrolytic reaction the N-acetylglucosamine glycosidic bond of metabolite 1. The reactions catalyzed by both the lytic glycosylases and NagZ are believed to involve oxocarbenium transition species. We describe herein the synthesis and evaluation of four iminosaccharides as possible mimetics of the oxocarbenium species, and we disclose one as a potent (compound 3, Ki = 300 A± 15 nM) competitive inhibitor of NagZ.

The use of beta-galactosidases from five different sources in syntheses through transglycosylation leads to 3-O-methyl allolactose, N-acetyllactosamine, N-acetylallolactosamine and Gal(beta1-6)Gal. The yield and distribution of products depends on the source of the enzyme and on the reaction conditions, viz. the nature of added organic co-solvent. Yields of 3-O-methyl allolactose (47%), Gal(beta1-6)Gal (6%), N-acetylallolactosamine (30%) were obtained in aqueous buffer with beta-galactosidase from E. coli. The same reactions occurred at much lower rates in the presence of water-miscible organic solvents. With beta-galactosidases from K. fragilis and A. oryzae, however, the synthesis of the above disaccharides occurred only in the presence of organic solvent (> 60% v/v triethyl phosphate, trimethyl phosphate or tetraglyme) but not in aqueous buffer solution. beta-galactosidases from D. pneumoniae and B. circulans in systems incorporating organic solvent produced 3-O-methyl allolactose and N-acetyllactosamine in yields of 30-40%. The direct separation of oligosaccharides on a preparative scale can be achieved by Ca2+-ligand exchange chromatography. Ultrafiltration was also used for the efficient recycling of enzymes.

Two simple regiospecific methodologies based on triazolic ring construction in the course of synthesis were applied for the synthesis of 1,2,3-triazolic nucleoside analogues. The cycloaddition reactions between diazomalonaldehyde and appropriate glycosylamine derivatives were rather effective, producing the desired nucleosides 11, 17 and 24. Diazotization of enamines 21a and 21b led to the corresponding triazolic ribonucleoside derivatives 22a and 22b, in good yields. Deprotection reaction of 22a, 22b and 24 was easily achieved by Lewis acid catalysis, producing the corresponding ribonucleosides 23a, 23b and 25.

The structural rationale, synthesis and evaluation of an inhibitor designed to block glucosamine synthesis by competitively inhibiting the action of glutamine: fructose-6-phosphate amidotransferase and subsequently reducing the transformation of any glucosamine-6-phosphate formed to UDP-N-acetylglucosamine are described. The inhibitor 2-acetamido-2,6-dideoxy-6-sulfo-d-glucose (d-glucosamine-6-sulfonate) is an analog of glucosamine-6-phosphate in which the phosphate group in the latter is replaced with a sulfonic acid group. The inhibitor is designed to function by three different modes which together reduce UDP-N-acetylglucosamine synthesis. This reduction was confirmed by evaluating the effect of the inhibitor on bacterial cell-wall synthesis and by demonstrating that it inhibits acetylation of glucosamine-6-phosphate competitively and by acting as a surrogate substrate. Inhibition of glucosamine production or suitably activated glucosamine in bacteria leads to disruption of the peptidoglycan structure, which results in softening, bulging, deformation, fragility and lysis of the cells. These modifications were documented by scanning electron microscopy for bacteria treated with the inhibitor. They were observed for inhibitor concentrations in the 20 mg/mL range for Escherichia coli and Bacillus subtilis and the 5 mg/mL range for Rhizobium trifolii.

A library of GlcNAc 6- or 1-phosphate analogues was designed, and each compound was evaluated computationally through docking studies for its binding affinity to AGM1/PGM3. The compounds with the highest binding affinity, as ranked through a docking score, were synthesised and screened for their ability to inhibit the production of UDP-GlcNAc. A glycofused oxazoline analogue showed good inhibition, and gave significant results in vitro.
