Celecoxib (CLX) is an anti-inflammatory drug that is used for acute pain treatment, but it has very low oral bioavailability owing to its poor water solubility. In this study, CLX solid dispersions are prepared by spray-drying using hydrophilic carriers with the aim of improving its apparent solubility and dissolution rate. Blends with different ratios of CLX, isomalt (ISO) and PVP were prepared in ethanol, and solid dispersions were obtained using a spray dryer. The saturation solubility and dissolution kinetics in 0.25% SLS and 0.04 M Na3PO4 containing media were determined. Also, SEM, DSC, XRPD, and stability studies were used to characterize the systems. Physicochemical analysis demonstrated the presence of amorphous CLX in the spray dried samples. The saturation solubility and dissolution rate of CLX from these formulations were higher than those for pure celecoxib and its physical mixtures. Amorphous CLX showed a lower dissolution rate compared to its crystalline form in 0.25% SLS medium, while this tendency was reversed under alkaline conditions. The CLX:PVP:ISO 3:5:2 spray dried sample showed the highest dissolution rate in both media. Exposure of samples to high moisture (75% humidity) recrystallized some of the amorphous CLX. Thus, the results showed that the dissolution rate of CLX was enhanced in 0.25% SLS, whereas a reduction in dissolution rate was observed in 0.04 M Na3PO4.

We describe a novel immobilization method that has been developed to overcome disadvantages of current immobilization techniques and to meet todays industrial requirements. Current methods are primarily very process specific and often suffer from scale-up limitations, thus preventing their industrial use. We demonstrate, that industrially already established processes can be replaced by cost-effective formulation technologies comprising of only a few process steps for producing the heterogeneous biocatalyst. Feasibility studies simulating industrial process conditions show that the granules provide good activity and sucrose conversion enabling their application in a fixed-bed reactor.

Effect of isomalt as novel binding agent on compressibility of poorly compactable paracetamol evaluated by factorial design

Tablets containing paracetamol were prepared by wet granulation. The model drug, paracetamol (API) and the diluent, milled isomalt (IM) were applied in different ratios where the granulation liquid was kept constant in each sample. The purpose of the study was to investigate the effect of wet granulation with milled isomalt on the tabletting properties of poorly compressible paracetamol. The results of tabletting process and the tablet characteristics were evaluated with 32 type face-centered full factorial design. The API:IM ratio (X1) and compression force (X2) were selected as independent variables. The dependent variables were as follows: the parameters of the tabletting process – lubrication coefficient (R-value), relative elasticity, friction work – and characteristics of the dosage form such as tablet hardness, friability, disintegration time, tensile strength. The results indicated that milled isomalt has an advantageous effect on both the process of compression and the tablet characteristics – due to its binder properties. Furthermore an optimal composition of the API and the diluent is to be found according to the compression process and the dosage form.

Celecoxib is a COX II inhibitor NSAID which is used for joint pains, rheumatoid arthritis and osteoarthritis, however due to its poor water solubility it shows very low oral bioavailability. Using solid dispersion formulations is one of the most promising strategies to increase solubility of poorly water soluble drugs. The purpose of this study is dissolution enhancement of celecoxib by preparation of solid dispersions via spray drying technique using PVP and Isomalt as hydrophilic carriers. Different ratios of celecoxib, Isomalt and PVP K30 (7:3:0, 5:5:0, 3:7:0, 1:9:0 and 3:5:2, 3:2:5) were prepared from 2% hydroalcoholic solutions (70:30 ethanol:water) using spray drier. Particle size analyzing, saturation solubility, SEM, DSC, FT-IR, XRPD and dissolution studies in 0.25% SDS and 0.04?M Na3HPO4 mediums were performed. Stability of samples was also studied after a week and a month storage at 75% humidity condition. The results showed that the saturation solubility of celecoxib in solid dispersion samples is 20?30 folds higher than raw celecoxib. Similar results have been shown for dissolution studies. Solid state analyses showed glass solution state of celecoxib in PVP/Isomalt matrixes. FTIR studies exhibited the formation of hydrogen bonding between celecoxib and PVP in these samples. Spray dried celecoxib (amorphous celecoxib) without usage of carrier showed lower dissolution rate compare to its crystalline state (in 0.25% SDS dissolution medium) whilst these results is vise versa in Na3PO4 dissolution medium. Interestingly almost all samples exhibited higher dissolution rate (in 0.25% SDS) after storage in 75% humidity. XRPD analysis demonstrated the crystallization of amorphous celecoxib after 1?month storage. In general using PVP K30 and Isomalt as hydrophilic carriers could increase solubility and dissolution rate of celecoxib in solid dispersion formulations.

The objective of this study was to produce a frozen yogurt containing low fat and no added sugar. Samples containing 5% polydextrose, 0.065% aspartame and acesulfame-K mixture, and different levels of inulin and isomalt (5.0, 6.5, and 8.0%) were produced at pilot scale and analyzed for their physical and chemical properties including proximate composition, viscosity, acidity, overrun, melting rate, heat shock stability, as well as sensory characteristics, and viability of lactic acid bacteria. With the addition of inulin and isomalt, viscosity increased by 19 to 52% compared with that of sample B (reduced-fat control). The average calorie values of samples substituted with sweeteners were about 43% lower than that of original sample. Low-calorie frozen yogurt samples melted about 33 to 48% slower than the reduced-fat control sample at 45 min. Based on quantitative descriptive profile test results, statistically significant differences among products were observed for hardness, iciness, foamy melting, whey separation, and sweetness characteristics. The results of principal component analysis showed that the sensory properties of the sample containing 6.5% inulin and 6.5% isomalt were similar to those of control. Lactic acid bacteria counts of frozen yogurt were found to be between 8.12 and 8.49 log values, 3 mo after the production. The overall results showed that it is possible to produce an attractive frozen yogurt product with the incorporation of inulin and isomalt with no added sugar and reduced fat.

Background: Twelves categories of active ingredients have been recognised to enhance human health. They are to some extent susceptible to certain conditions such as heat, light and low pH. To reduce their susceptibility and achieve controlled release at the target site, various microencapsulation strategies have been introduced. Scope and approach: In this review, the chemical structures, physicochemical properties and beneficial effects of the active components are summarised. Different encapsulation techniques and tailored shell materials have been investigated to optimise the functional properties of microcapsules. Several encapsulated constituents (e.g., amino acids) have been successfully incorporated into food products while others such as lactic acid bacteria are mostly used in the free format. Encapsulating some of these active ingredients will extend their ability to withstand process conditions such as heat and shear, and prolong their shelf stability. Key findings and conclusions: The functional properties of a microcapsule are encapsulation efficiency, size, morphology, stability, and release characteristics. Several microencapsulation strategies include the use of double emulsions, hybrid wall materials and crosslinkers, increasing intermolecular attraction between shell and core, physical shielding of shell materials, and the addition of certain ions. Other approaches such as the use of hardening agents, nanoencapsulation, or secondary core materials, and the choice of shell materials possessing specific interactions with the core may be used to achieve targeted release of active ingredients. The physicochemical properties of shell materials influence where the active ingredients will be released in vivo. A suitable microencapsulation strategy of active ingredients will therefore expand their applications in the functional foods industry.

The current study determines optical and fluorescence response of halogen substituted series of meso-tetrakis-(4-halophenyl) porphyrin; H2TXPP (Halo = F, Cl, Br, I) dye in tetrahydrofuran; THF and THF-water system at changing pH in relationship with changing medium of allure. Effects produced by varying the pH and medium, over spectral and aggregation were discussed in detail. Results show sequential protonation and deprotonation of H2TXPP series in acidic (pH = 4) and (pH =10) basic medium. Specific structural changes of monomeric absorption band were put in evidence on lowering pH, which includes broadening and splitting of soret or B band. Other changes include increasing in intensity and red-shifting of Q1 band indicating some degree of aggregation. The side-by-side aggregation and formation of J-aggregate were quite evident.
