beta-N-Acetylhexosaminidase (EC 3.2.1.52) was purified from the culture filtrate of Pycnoporus cinnabarinus to homogeneity by polyacrylamide disc gel electrophoresis. The ratio of beta-GlcNAcase activity to beta-GalNAcase activity remained constant during the purification process. The molecular weight of this enzyme was estimated to be about 120,000 by gel filtration, and the isoelectric point was at about pH 5.4.The optimum pH was at 2.2 for pNPGlcNAc and around 3.7 for pNPGalNAc.The enzyme was relatively stable at acid pH range of 2-4 (for 45 hr at 5 deg C) and below 45 deg C (for 10 min at pH 2.8).The enzyme hydrolysed chito-oligosaccharides, such as N,N-diacetylchitobiose, N,N,N-triacetylchitotriose and N,N,N,N-tetraacetylchitotetraose.

Elevated hyaluronan expression is a hallmark of many types of cancer. Therefore, inhibition of hyaluronan biosynthesis can potentially slow the growth of tumor cells. Herein, we explore a chain termination strategy to reduce hyaluronan synthesis by tumor cells. Several analogs of glucosamine were prepared, which contained modifications at the C-3 positions. These analogs can possibly cap the nonreducing end of a growing hyaluronan chain, thus lowering the amount of hyaluronan synthesized. Upon incubation with pancreatic cancer cells, a fluorine-containing glucosamine analog was found to exhibit significant inhibitory activities of hyaluronan synthesis. Furthermore, it drastically reduced the proliferation of cancer cells. Supplemental materials are available for this article. The numerous biological roles of LacNAc-based oligosaccharides have led to an increased demand for these structures for biological studies. In this report, an efficient route for the synthesis of beta-galactosides using a bacterial beta-4-galactosyltransferase/-UDP-4?-gal-epimerase fusion protein is described. The lgtB gene from Neisseria meningitidis and the galE gene from Streptococcus thermophilus were fused and cloned into an expression vector pCW. The fusion protein transfers galactose to a variety of different glucose- and glucosamine-containing acceptors, and utilizes either UDP-galactose or UDP-glucose as donor substrates. A crude lysate from Escherichia coli expressing the fusion protein is demonstrated to be sufficient for the efficient preparation of galactosylated oligosaccharides from inexpensive UDP-glucose in a multigram scale. Lysates containing the fusion protein are also found to be useful in the production of more complex oligosaccharides in coupled reaction mixtures, e.g., in the preparation of sialosides from N-acetylglucosamine. Thus, bacterially expressed fusion protein is well suited for the facile and economic preparation of natural oligosaccharides and synthetic derivatives based on the lactosamine core.

The CB2 receptor is an attractive target for the treatment of a wide range of diseases and pathological conditions. Compounds that selectively activate the CB2 receptor are desirable as this avoids CB1-mediated psychoactive effects. Heteroarylidene-benzamides have demonstrated efficacy as selective CB2 receptor agonists. We aimed to expand the structure-activity relationship studies of this series of compounds by investigating the heteroaromatic core via the synthesis and in vitro evaluation of a small library of various heteroaromatic benzamide analogues. As heteroaromatic amides are privileged scaffolds in drug design, methods to synthesise them are of interest. Concise and reliable synthetic strategies were developed to access these novel analogues. The ?ylidene-benzamide moiety is shown to be essential for CB activity as all amide derivatives exhibit no functional activity at either CB2 or CB1 receptors.

A series of 5-alkenyl and 6-alkenyl aldehydes bearing aryl and ester functionality on the alkene and with and without 1,3-dioxolane substitution at the 3-position has been prepared. The N-methyl and N-benzyl nitrones derived by the condensation of the aldehydes with the corresponding hydroxylamines undergo intramolecular 1,3-dipolar cycloaddition on heating (or at room temperature in two instances) to give either bridged or fused isoxazolidines depending on the regiochemistry of the cycloaddition reaction. The 6-alkenyl nitrones give fused products in all cases except those bearing an aryl group at C-6, which give bridged products. All of the 5-alkenyl nitrones prepared in this study cyclized to afford fused isoxazolidines.

COA of Formula: C10H13N5O, Some examples of the diverse research done by chemistry experts include discovery of new medicines and vaccines, improving understanding of environmental issues, and development of new chemical products and materials. An article , which mentions 7306-67-4, molecular formula is C10H13N5O. The compound – 9-(Tetrahydro-2H-pyran-2-yl)-9H-purin-6-amine played an important role in people’s production and life.

Uracil derivatives form strong complexes with complementary 2,4-diaminotriazine and adenine compounds, whereas derivatives of 5-azauracil (2,4-dioxotriazine) are known to form weak complexes in aqueous medium. However, herein we report that in organic medium (CDCl3), the 5-azauracil moiety forms hydrogen-bond-mediated complexes with complementary 2,4-diaminotriazine and adenine compounds, with strengths comparable to those formed by uracil compounds. Such dichotomous base-pairing behavior of the 5-azauracil moiety, in organic versus aqueous media, is found to be consistent with the ionization of the 5-azauracil moiety in aqueous medium leading to competitive interference from water molecules (via solvation), which is absent (lack of such ionization and solvent interference) in organic medium. This discriminating role of solvent (e.g., water) could have been an important factor in the selection of molecules, based on their physicochemical properties, and subsequently in the emergence of potential primordial informational oligomers that would have played a role in the origins of life.

C-glycosides are important compounds as they are used as bioactive molecules and building blocks. We have developed methods to concisely synthesize C-glycosides from unprotected 2-N-acyl-aldohexoses and unactivated ketones; we designed aldol-condensation-oxa-Michael addition reactions catalyzed by amine-based catalysts using additives. Depending on the conditions used, C-glycosides were stereoselectively obtained. Our methods allowed the C-C bond formations at the anomeric centers of unprotected carbohydrates under mild conditions to lead the C-glycosides in atom- and step-economical ways.
